Mechanism of action

Kisspeptin-10 is a 10-amino acid C-terminal fragment of kisspeptin (also called metastin), the endogenous ligand for the KISS1R receptor. The kisspeptin-GnRH axis is the master controller of the hypothalamic-pituitary-gonadal (HPG) axis — kisspeptin neurons integrate metabolic status, stress, and reproductive signals to regulate GnRH pulse frequency and amplitude. Kisspeptin-10 (the minimal bioactive fragment) activates KISS1R to trigger GnRH release, which drives LH and FSH secretion, and downstream testosterone/oestradiol production. In hypogonadotrophic hypogonadism (low LH/FSH with normal gonads), kisspeptin stimulation can restore the hormonal axis. Human Phase I/II trials demonstrate robust LH and testosterone responses.

Documented effects (research-model)

  • Robust LH and FSH stimulation in human trials
  • Testosterone restoration in hypogonadotrophic hypogonadism
  • Ovulation induction research (women)
  • HPG axis restoration post-steroid use
  • Sexual desire enhancement via testosterone axis
  • Hypothalamic amenorrhoea treatment research

Synergies

  • PT-141 MODERATE
Kisspeptin elevates testosterone (hormonal desire); PT-141 activates central arousal pathways. Comprehensive sexual health protocol.
  • Gonadorelin HIGH
Both stimulate the HPG axis at different levels — kisspeptin upstream of GnRH, gonadorelin as GnRH itself. Often combined in clinical HRT adjunct protocols.

Myths & misconceptions

Myth Kisspeptin-10 is just another testosterone booster supplement.
Reality Kisspeptin-10 is a research peptide with documented human pharmacodynamic data showing genuine LH and testosterone elevation. It is not comparable to herbal "testosterone boosters" — it acts on the central HPG axis with measurable hormonal effects in clinical studies.

Evidence gaps

  • Optimal dosing protocol for sustained hormonal restoration not established.
  • Long-term receptor desensitisation with sustained use not fully characterised.
  • Efficacy in the context of exogenous testosterone use (HPG axis suppressed) not well studied.

Safety notes

Well-tolerated in Phase I/II trials. Transient injection site reactions. Monitor LH, FSH, testosterone, and oestradiol during research. Pulsatile administration may be more physiological than bolus dosing. Research context only.
