Mechanism of action
5-Amino-1MQ is a small molecule (not technically a peptide) inhibitor of NNMT (Nicotinamide N-methyltransferase) — an enzyme that uses the methyl donor SAM to methylate nicotinamide, depleting the SAM pool and diverting nicotinamide away from NAD+ synthesis. By inhibiting NNMT, 5-Amino-1MQ preserves nicotinamide for NAD+ biosynthesis (via NAMPT) and maintains the SAM pool for methylation reactions. In animal models, it reduced adipocyte size, improved insulin sensitivity, and elevated NAD+ levels without the side effects associated with high-dose NMN/NR supplementation.
