Mechanism of action

Pinealon is a synthetic tetrapeptide (Ala-Glu-Asp-Gly) from the Khavinson peptide bioregulator series, derived from the bovine pineal gland. Like Epitalon (from the same Khavinson research group), Pinealon exerts gene-regulatory effects by binding directly to DNA promoter regions and modulating transcription of genes involved in antioxidant defence, circadian rhythm regulation, and neuroprotection. It has been studied for improving cognitive function in elderly subjects, reducing neuronal damage in models of ischaemia and hypoxia, and regulating melatonin-related gene expression in the pineal gland. The primary hypothesis is that age-related pineal degeneration contributes to neurological decline, and Pinealon partially compensates for this.

Documented effects (research-model)

  • Neuroprotection via gene expression modulation
  • Circadian rhythm and sleep architecture support
  • Antioxidant enzyme upregulation in neural tissue
  • Cognitive protection in ageing models
  • Potential synergy with melatonin signalling

Synergies

  • Epitalon HIGH
Epitalon (pineal/telomere-focused) + Pinealon (pineal/neuroprotection). Both from Khavinson series — designed to be used together in longevity protocols.
  • DSIP MODERATE
Both influence sleep and circadian biology. Pinealon (pineal regulation) + DSIP (delta sleep induction). Complementary sleep-focused stack.

Myths & misconceptions

Myth Pinealon is a reliable melatonin booster.
Reality Pinealon modulates pineal gland gene expression — it does not directly elevate melatonin in a dose-dependent, predictable way. Its effects on circadian biology are indirect and not well quantified in humans.
Myth Pinealon has the same evidence as Epitalon.
Reality Pinealon has considerably less published evidence than Epitalon. Epitalon has 40+ publications including some human data; Pinealon research is much more limited.

Evidence gaps

  • Minimal published human research — almost all data from Russian-language animal and in vitro studies.
  • Optimal cycle length and long-term effects not established.
  • Mechanism of neuroprotection not definitively characterised in humans.

Safety notes

Very limited human safety data. Based on the Khavinson bioregulator safety profile, short cycles (10 days) with long washout periods (4 months) are the standard protocol. Research context only.
